Serisin kaplı altın nanoparçacık sentezlenmesi ve karakterizasyonu

Abstract

Bu çalışmada, Bombyx mori ipek böceği kozasından elde edilen ipek serisini kullanılarak serisin kaplı altın nanoparçacıklar (S-AuNP) sentezlenmiştir. Çözeltideki altın tuzu miktarı (10 mL, 1 mM) sabit tutularak buna farklı derişimlerde (%0,25, 0,5 ve 1) ve pH değerlerinde (8 ve 12) hazırlanmış 10 mL’lik serisin çözeltilerinden eklenerek deney grupları oluşturulmuştur (S-AuNP0,25, S-AuNP0,5, S-AuNP1). Bütün S-AuNP gruplarındaki kırmızı renk değişimi ve 520-530 nm dalga boylarında gözlemlenen belirgin yüzey plazmon rezonans (SPR) tepeleri nanoparçacık sentezlendiğinin göstergesidir. Ancak, bu kırmızı renk değişimi ve tepe noktaları pH 12’de belirgin değildir. FTIR analizine göre, serisinde bulunan N-H, O-H ve C=O fonksiyonel gruplarının indirgeme reaksiyonunda görev aldıkları düşünülmektedir. TEM görüntülerinde, artan serisin derişimiyle ortalama parçacık boyutu ve boyut dağılımı büyümüştür. Serisin nanoparçacıkların yüzey yükünü negatif yapmıştır ve S-AuNP0,5 ve S-AuNP1 gruplarının negatif zeta potansiyelinin artması suda S-AuNP0,25 grubuna göre daha kararlı olduklarını göstermektedir. Bu bulgu, tuzda kararlılık deneyleriyle de teyit edilmiştir. S-AuNP0,25, 1 günlük inkübasyon periyodu sonucunda sıvıdaki kararlılığını yitirmiş ancak diğer gruplar 7 gün boyunca kararlı kalmıştır. Bütün sonuçlar değerlendirildiğinde, en uygun grubun S-AuNP0,5 olduğu kabul edilmiştir. Bu şekilde üretilen nanoparçacıkların biyomedikal sektöründe nanoboyutta ilaç taşıyıcı sistemler olma vb gibi potansiyel uygulama alanları olabilir.
