Alkol Verilen Sıçanlarda 2,4-Diklorofenoksi Asetik Asitin Hematolojik Parametreler Üzerine Etkilerinin Belirlenmesi

Abstract

Modern tarımda, pestisit, herbisit ve çeşitli gübreler gibi birçok bileşik, bitki üretimini arttırmak için sıklıkla kullanılmaktadır. Bununla birlikte, bu bileşikler arasında, sentetik herbisitlerin tarımsal amaçlı yaygın kullanımı, canlı organizmalar üzerinde olumsuz bir etkiye sahip olup, insan ve hayvanlarda çeşitli hematolojik ve nörolojik komplikasyonlara yol açabilmektedir. Alkol tüketimi, kardiyovasküler hastalıkta potansiyel risk faktörlerinden biridir. Bu çalışmanın amacı, yaygın olarak çiftçiler tarafından herbisit olarak kullanılan 2,4-diklorofenoksi asetik asidin (2.4-D), etil alkol verilen sıçanlarda hematolojik değişikliklere etkisini araştırmaktır. Toplam 28 Sprague Dawley erkek sıçan rastgele olarak grup başına 7 hayvan içerecek şekilde dört gruba ayrıldı. Birinci grup kontrol olarak görev yaparken, deney grupları şöyledir; herbisit grubu (5 mg / kg, 2,4 D, oral yoldan), alkol grubu (3 mg / kg etil alkol, oral yoldan), herbisit + alkol grubu (5 mg / kg 2,4D ve 3 mg / kg etil alkol, oral yoldan).  Altmış günlük toplam deney süresi sonunda kan örnekleri anestezi altında kalp punksiyonu ile hayvanlardan alındı. Kan örneklerinden eritrosit (RBC), toplam lökosit sayısı (WBC)  ve yüzde oranları ile trombosit sayısı (PLT), hematokrit değeri (HCT) ve hemoglobin (HGB) düzeyi belirlendi. 2,4-D uygulanan hayvanlarda, WBC, lenfosit, monosit ve granülosit yüzdelik değerlerinin arttığı bulundu. Alkol ve alkol ile birlikte 2,4-D uygulanan hayvanlarda, RBC, HGB, HCT ve PLT sayılarının azaldığı tespit edildi. Alkol, 2.4-D ve alkol ile birlikte 2.4 -D uygulamalarının, her bir kırmızı kan hücresindeki ortalama hemoglobin seviyesini (MCH) arttırdığı gözlendi. Sonuç olarak, alkol verilen sıçanlarda 2,4-D uygulamasının hematolojik parametrelerde değişikliklere neden olduğu sonucuna varıldı.

Determination of The Effects of 2,4-Dichlorophenoxy Acetic Acid on Hematological Parameters in Rats Given Alcohol

Abstract

In modern agriculture, many compounds such as pesticides, herbicides and various kinds of fertilizers are frequently used in the fields to increase crop production. However, among these compounds, widespread usage of synthetic herbicides for agricultural purpose has an adverse effect on living organisms and can lead to several hematological and neurological complications in human and animals. Alcohol consumption is one of the potential risk factors in cardiovascular disease. The aim of this study is to investigate the effects of 2,4-dichlorophenoxy acetic acid (2.4-D), used commonly by farmers as a herbicide, on hematological changes in rats given alcohol. Totally 28 Sprague Dawley male rats were randomly divided into four groups containing 7 animals per group. While first group served as a control, experimental groups were as follows; herbicide group (5 mg/kg, 2,4- D, orally), alcohol group (3 mg/kg ethyl alcohol, orally), herbicide plus alcohol group (5 mg/kg 2,4-D and 3 mg/kg ethyl alcohol, orally). In the end of experimental period lasted totally 60 days, the blood samples were taken from animals by cardiac puncture under anesthesia. In the blood samples, erythrocyte (RBC), total leukocytes (WBC) and percentage of the different white blood cells, hematocrit value (HCT), hemoglobin concentration (HGB) and platelet counts (PLT) were determined. 2,4-D increased the WBC,  percentage of lymphocyte, monocyte and granulocyte numbers. Alcohol and alcohol plus 2,4-D application reduced RBC numbers, HGB, HCT value and PLT concentration. The mean corpuscular hemoglobin level (MCH) was increased by alcohol, 2.4 D and alcohol plus 2.4 D treatments. We concluded that 2,4-D application in rats given alcohol leads to changes in hematological parameters.
