Gebelik esnasýnda diazinon’a maruz kalan ratlarýn yenidoðan yavrularýnýn akciðer dokusunda oluþan morfolojik deðiþiklikler ve bu deðiþikliklere kafeik asit fenetil ester’in etkisinin araþtýrýlmasý

Öz

Objectives: The aim of present study is to investigate morphological changes occurred in lungs of newborn rats whose mothers exposed to diazinon during their pregnancy, and effects of CAPE on these changes. Methods: Thirty female Wistar albino rats were randomly divided into three groups, as follows: Group I(n; 10): control group, Group II (n; 10): 25mg/kg/day diazinon treated group in the 14-18 days of pregnancy, Group III (n; 10): 25 mg/kg/day diazinon + 10 µmol /kg/day CAPE treated group in the 14-18 days of pregnancy. Diazinon administrated by oral gavage in corn oil and CAPE administrated intraperitoneally. Results: In group II, diazinon causes low weight gain in pregnant rats and low birth weights in their newborns. Histopathological examinations of lung tissues revealed intraparenchymal inflammatory infiltration, alveolar and bronchial hemoraji, intraparenchymal vascular congestion and thrombozis and emphysematous changes. In group III, administration of CAPE has considerably reduced these pathological changes. As a result, low weightgain in pregnant rats and low birth weights in their newborns also, histopathological changes in the lung tissues of newborn rats determined. It is also concluded that CAPE has ameliorating effects on these histopathological changes.

Öz

Amaç: çalışmanın amacı gebelikte diazinon'a maruz kalan bebe ratların yenidoğan yavrularının akciğer dokusunda oluşan morfolojik değişiklikleri ve bu değişikliklere kafeik asit fenetil ester'in (CAPE) nin etkisinin araştırılmasıdır. Gereç ve Yöntem: Bu çalışmada 30 Wistar albino rat kullanıldı. Ratlar rastgele 3 gruba ayrıldı. Grup 1: kontrol grubu (n;10), grup 2 (n;10): Gebeliğin 14-18. günlerinde 25 mg/kg/gün diazinon verilen grup, grup 3 (n;10): Gebeliğin 14-18. günlerinde 25 mg/kg/gün diazinon + 10 µmol /kg/day CAPE verilen grup. Deney gruplarında diazinon oral gavaj ile CAPE ise intraperitoneal uygulandı. Gebe ratların gebelik boyunca kilo alımları ve yenidoğan yavruların doğum ağırlıkları takip edildi. Yenidoğan yavrular 3. günde dekapite edildi. Akciğerleri diseke edilerek %10'luk formaldehid içerisine alınarak rutin histalojik doku takibi yapıldı ve preperatlar ışık mikoskobunda değerlendirildi. Bulgular: Grup 2'deki anne ratların ve yenidoğanlarının ortalama kiloları kontrol grubuna göre daha düşüktü. Ayrıca grup 2'deki yenidoğanların akciğerlerinde intraparankimal inflamatuar infiltrasyon, alveolar ve bronşiolar hemoraji, intraparankimal vasküler konjesyon ve trombosis, ve amfizematöz değişiklikler gözlendi. Diazinon ve CAPE'nn birlikte verildiği grup 3'te ise bu patolojilerin önemli oranda azaldığı tespit edildi. Sonuç olarak gebelikte diazinon'a maruz kalan gebe ratların kilo alımlarında ve yeni doğanlarının kilolarında azalma ayrıca akciğer dokularında histopatolojik değişiklikler tespit edildi ve CAPE uygulanması ile bu değişikliklerin önemli derecede azaldığı gözlendi.
