Investigation of Neospora caninum and Toxoplasma gondii tachyzoites in peripheral blood samples ofdogs by TaqMan probe based Real Time PCR

Abstract

This study was carried out to investigate Neospora caninum and Toxoplasma gondii tachyzoites in total of 400 whole peripheral blood samples with EDTA which were collected from the clinically healthy dogs in Kayseri region. Following the isolation of genomic DNA from the blood samples, spectrophotometric measurements of the obtained genomic DNAs were performed and appropriate concentrations were prepared for Real Time PCR (qPCR). qPCR analyses of the samples were conducted using the specific primers and probes that target the gene regions NC5 for N. caninum and RE for T. gondii. Out of 400 blood samples, 15 (3.8%) were found to be infected with N. caninum while no T. gondii positivity was determined in the examined samples. In conclusion, some molecular epidemiological data about N. caninum and T. gondii infections dogs in Kayseri region were firstly demonstrated in Turkey with this study

Köpek periferal kan örneklerinde Neospora caninum ve Toxoplasma gondii takizoitlerinin TaqMan prob bazlı Real Time PCR ile araştırılması

Abstract

Bu çalışma, Kayseri yöresinde klinik olarak normal görünümlü köpeklerden toplanmış toplam 400 adet EDTA'lı periferal tam kan örneğinde Neospora caninum ve Toxoplasma gondii takizoitlerinin araştırılması amacıyla yapılmıştır. Kan örneklerinden genomik DNA izolasyonunu takiben, elde edilen genomik DNA’ların spektrofotometrik ölçümleri yapılmış ve Real Time PCR (qPCR) için uygun konsantrasyonlar hazırlanmıştır. N. caninum için NC5 ve T.gondii için RE gen bölgelerini hedef alan spesifik primer ve problar kullanılarak örneklerin qPCR analizleri gerçekleştirilmiştir. qPCR sonuçlarına göre 400 örneğin 15’inde (%3,8) N. caninum pozitifliği tespit edilmiş olup hiçbir örnekte T. gondii pozitifliğine rastlanmamıştır. Sonuç olarak bu çalışma ile Türkiye’de ilk kez Kayseri yöresinde köpeklerin perifer tam kan örneklerinde N.caninum ve T. gondii takizoitleri TaqMan Prob Bazlı Real Time PCR tekniği ile araştırılmış ve moleküler epidemiyolojik veriler elde edilmiştir
