Pathological findings of experimental Candida albicans infection in guinea pigs

Abstract

Experimental candidiasis was performed via different routes in guinea pigs immunosuppressed with cylophosphamide and dexamethasone by administrating C. albicans ATCC 26555 serotype A strain. Three experimental groups were constituted by administrating agent through intravenous (Group I, n: 12), intraperitoneal (Group 2, n: 12) and oral (group 3, n: i2) routes. Four guinea pigs for eaeh group were used to set up control (Group 4, n: i2) and physiologic saline solution was injected same routes as experimental groups. The guinea pigs have died in Group i and Group 2, at the percent of 58,3 % and 41,6 %, respectively. However, mortality was not observed in Group 3. The lesions were different in location depending on the inoculation routes. Macroscopieally, multifocal gray to white foci were observed intensively in the lung,liver, heart, spleen and kidney in Group 1. In Group 2, the macroscopical lesions were generally seen in the abdominal cavity organs and testicles. However, in Group 3, the lesions were observed usually on the upper digestive tracts. Microscopically, systemic infection characterized by pyogranulomatous lesions was observed in Group i and Group 2. In addition the similar lesions were seen in the testicles of Group 2. In Group 3, local infection characterized by hyperkeratosis of the squamous epithelium and uleers covered with pseudomembranes was occuned. Histochemically, blastospores, pseudohyphae and hyphae were see n in the all lesions.

Kobaylarda deneyselCandida albicans enfeksiyonunda patolojik bulgular

Abstract

Çalışmada, dekzametazon ve siklofosfamid ile bağışıklık sistemi baskılanan erkek kobaylara farklı yollarla C. albicans serotip A (ATCC 26555) suşu verilerek deneysel kandidiazis oluşturuldu. Hayvanlar intravenöz (Grup i,n: 12), intraperitoneal (Grup 2, n:12) ve oral (Grup 3, n:12) yolla enfekte edildi. Kontrololarak (Grup 4, n:12) her deney grubu için 4 kobaya aynı yollarla fizyolojik tuzlu su verildi. İnokülasyon sonrası kobayların Grup i'de % 58,3 'ü, Grup 2'de % 41 ,6'sl öldü. Buna karşın Grup 3'teki kobaylarda ölüm şekillenmedi. Enfeksiyonda şekillenen lezyonların dağılımı etkenin veriliş yoluna göre farklılık gösterdi. Makroskobik olarak Grup i'de çoğunlukla akciğer, karaciğer, kalp, dalak ve böbrekte çok sayıda boz beyaz renkte odaklar şekillendi. Grup 2'de makroskobik lezyonlara çoğunlukla karın boşluğu organları ve testislerde rastlandı. Grup 3 'te ise lezyonlar çoğunlukla üst sindirim sisteminde dikkati çekti. Histopatolojik olarak Grup i ve 2'de çeşitli organlarda pyogranülomatöz lezyonlarla karakterize sistemik enfeksiyon oluştu. Ayrıca, Grup 2'de testislerde de lezyonlara rastlandı. Grup 3'te ise çoğunlukla üst sindirim sisteminde keratinize epitelde keratin fazlalaşması ve üzeri yalancı membranla örtülü ülserlerIe karakterize yersel enfeksiyon gözlendi. Histokimyasal olarak tüm lezyonlarda, blastospor, yalancı ve gerçek hifalar saptandı
