Immunohistochemical distribution of COX-1 and COX-2 in the liver to during the puberty period ofrats applied with capsaicin

Abstract

In this study, the effect of capsaicin on the immunohistochemical distribution of cyclooxygenase-1 (COX-1) and cyclooxygenase-2 (COX-2) were examined in liver of the rat at the pubertal age.Thirty rats (Sprague-Dawley) aging 50 days old were used in this study. Rats were divided into three groups; experimental, sham and control group. Experimental group (n = 10) rats, was injected 1 mg / kg doses of capsaicin (10% ethanol, 1% tween, containing 80% distilled water mixture) subcutaneously every day for a period of one week. Sham group (n = 10) rats, were subcutaneously injected with a mixture containing 10% ethanol and 1% tween within 80% distilled water. The control group made no application. A week after the last injection, rats of three groups body was weighed and euthanized with ether anesthesia and liver tissues were taken. Body weights compared within the groups.The tissue localization of COX-1 and COX-2 was analysed by immunohistochemical method.Streptavidin-biotin-peroxidase complex technique was applied.COX-1 immunoreactivity experimental, sham and control groups were observed in centralis vein and the environment Kiernan centrilobuler region. Immunoreactivity was found in similar concentrations in all three groups. COX-2 is a experimental, sham and control groups were observed in centralis vein and Kiernan centrilobuler region in the portal area

Puberte döneminde capsaicin uygulanan sıçanların karaciğerinde COX-1 ve COX-2' nin immunohistokimyasal lokalizasyonu

Abstract

Çalışmamızda, puberte döneminde capsaicin uygulanan sıçanların karaciğerinde siklooksijenaz-1 (COX-1) ve siklooksijenaz-2 (COX-2)’nin immunohistokimyasal lokalizasyonu incelendi. Çalışmada 50 günlük 30 erkek sıçan (Sprague-Dawley ırkı) kullanıldı. Sıçanlar deneme, sham ve kontrol olmak üzere 3 gruba ayrıldı. Deneme grubundaki (n=10) sıçanlara subkutan yolla her gün 1 mg/kg dozda capsaicin, % 10 ethanol, % 1 tween, % 80 distile su içeren karışım bir hafta süre ile enjekte edildi. Sham grubunu (n=10) oluşturan sıçanlara da aynı deneme grubundaki sıçanlarda olduğu gibi subkutan yolla CAP yerine sadece % 10 ethanol, % 1 tween, % 80 distile su içeren karışım subkutan olarak yapıldı. Kontrol grubuna (n=10) ise herhangi bir uygulama yapılmadı. Bir haftanın sonunda her üç gruptaki hayvanların vücut ağırlıkları tartıldıktan sonra eter ile ötenazi edildi ve karaciğer dokuları alındı. Grupların canlı ağırlıkları karşılaştırıldı. COX-1 ve COX-2 lokalizasyonu, immunohistokimyasal yöntemle tespit edildi. Streptavidin-Biotin-Peroksidaz Kompleks tekniği uygulandı. COX-1’in immunoreaktivitesi deneme, sham ve kontrol gruplarında vena sentralis ve Kiernan aralığı çevresindeki hepatositlerin sitoplazmasında gözlendi. Her üç grupta da immunoreaktivite birbirine benzer yoğunlukta bulundu. COX-2 ise deneme, sham ve kontrol gruplarında vena sentralisler etrafında ve Kiernan aralığına kadar olan portal alanda gözlemlendi
